This study aims to explore the molecular mechanism of neuronal axonal lesions caused by high glucose, and investigate protective effect Lycium barbarum polysaccharide (LBP) on axon damage with in vitro models. A rat model type II diabetic encephalopathy was established, rats were treated LBP for 10 weeks. Morris water maze experiment used detect spatial learning memory ability. Neuronal measured vivo neuron tracing experiment. Immunohistochemistry Western-blot technology employed protein localization expression. Combined primary culture, total internal reflection fluorescence microscopy effects glucose transport. PI3K inhibitors LY294002 an activator insulin signaling pathway possible mechanism. In experiments showed that hippocampus tissue STZ-induced rats, axons damaged, accompanied hyperphosphorylation Tau protein, which subsequently resulted transport cognition dysfunction. significantly reduced peripheral blood serum levels thereby alleviating resistance. At same time, can improve impairment brain pathology rats. treatment effectively block PI3K/Akt pathway. The phosphorylation level LY294002+ group is higher than intervention group, while reverse blocking LBPs. reduce up-regulating PI3K/Akt/GSK3β pathway, a damage, improving cognitive function performance
